DNS METHOD FOR ESTIMATION OF REDUCING SUGARS






Reducing sugars are the one that have a free aldehyde or ketone group
eg:- Glucose have an aldehyde group and fructose having ketone group.So they are reducing sugars.
Usually all the monosaccharides have reducing properties. However in Disaccharides,Sucrose is not a reducing sugar. Because sucrose is a composition of GLucose and Fructose linked together by 
α (1-2)Glycosidic bond,and it does not have free aldehyde or ketone group.

ESTIMATION OF REDUCING SUGARS BY DNS METHOD:-

DNS stands for 3,5 Dinitrosalicylic acid. When reducing sugars are treated with DNSA in boiling water bath for specific time(10 minutes at 100°C ), 3,5 DInitrosalicylic acid is converted into 3 Amino 5 Nitro Salicylic acid which has characteristic Yellow colour. Intensity is directly proportional to concentration, i.e., Higher the concentration, the intensity of the colour increases. The OD value for this experiment (Optical Density value) can be efficiently observed at 540 nm.Hence by measuring the OD value for known concentrations of the reducing sugar and plotting its value on the graph, a standard curve can be obtained . By using this, the concentration of unknown reducing sugar can be obtained
